Abnormal periostracum secretion during the mineralization process of the pearl in the blacklip pearl oyster Pinctada margaritifera ArchiMer

Whitish parasite-like bodies (PLBs), 0.5-2 mu m in size, were detected within the pearl bag of some grafted pearl oysters, Pinctada margaritifera, from Takapoto island (French Polynesia). Histological and ultrastructural investigations have revealed that these bodies consisted of lamellar organic material locally secreted by some epithelial cells of the pearl bag. The fine structure and the formation of these bodies are described. The causes of this abnormal secretion of organic mater, which actually corresponds to periostracum, are not presently known. No parasite nor infectious micro-organism was found associated with the PLBs.

Bilan des analyses du réseau microbiologique (REMI), en Languedoc-Roussillon, d'avril 1989 à décembre 1991 ArchiMer

Le réseau de surveillance microbiologique d'IFREMER a été mis en place dans toute la France en 1989, dans le but de garantir la santé publique et de suivre la qualité de l'environnement. Dans la région Languedoc-Roussillon sept sites producteurs de coquillages sont surveillés chaque mois ou plus fréquemment si· nécessaire. Dans le présent rapport, les méthodes d'analyses et les critères d'étude des données sont décrits; les résultats acquis d'avril 1989 à décembre 1991 sont présentés pour chaque site en essayant d'expliquer les contaminations observées par les sources de pollution potentielles recensées, la pluviométrie ou tout autre facteur local. A bloom of the toxic dinoflagellate Alexandrium tamarense was observed for the first time in Thau iagoon between November and December 1998. Maximum cell concentrations detected in shellfish farming areas (85 000 Cells/1) were high enough to make mussels and clams unsuitable for human consumption (max. 850 ug. STX eq.100 g'1) while toxic levels in oysters did not exceed, or even reach, the international public health threshold established for PSP toxins. Les microorganismes, bactéries et virus d'origine entérique, sont abondants dans les rejets urbains. Si la charge bactériologique contenue dans les eaux usées est notablement réduite lors des traitements appliqués dans les stations d'épurations, de nombreux microorganismes persistent et sont rejetés avec les eaux épurées dans le milieu récepteur, en rivière ou directement en mer. Le développement de méthodes analytiques basées sur la biologie moléculaire a permis récemment la détection de virus entériques non cultivables dans les échantillons de l'environnement, tels que les entérovirus, le virus de l'hépatite A, les calicivirus, les astrovirus et les rotavirus. Lagune vouée de longue date à la conchyliculture, l'étang de Thau fait l'objet d'une surveillance microbiologique régulière des coquillages issus du milieu et livrés à la consommation directe. La présente étude traite de la qualité bactériologique des coquillages observée de 1971 à 1998, période marquée par la restructuration et l'accroissement des implantations conchylicoles dans le cadre d'un plan qui a pris effet à partir de 1971. La présentation géographique du site est suivie d'un rappel historique des changements intervenus dans le dispositif de surveillance sanitaire en fonction de l'évolution des pratiques conchylicoles et de la situation du bassin versant. Sometimes, the monitoring network results provide an inhibited point of view to understand bacteriological process on the field. We must go further in contamination spatial or temporal structures to obtain a global picture. At Leucate, if a part of bacteriological contamination is correlated to precipitations, the main environmental factor to explain local variability seems to be the monthly variability population. This fact, completed by a spatial design, reveals a concentric gradient in the shellfish farrning area. Sorne hypothesis are discussed to explain this contamination (resurgence, aquatic birds droppings, ...)

Penaeidins, antimicrobial peptides with chitin-binding activity, are produced and stored in shrimp granulocytes and released after microbial challenge ArchiMer

Penaeidins are members of a new family of antimicrobial peptides isolated from a crustacean, which present both Gram-positive antibacterial and antifungal activities. We have studied the localization of synthesis and storage of penaeidins in the shrimp Penaeus vannamei. The distribution of penaeidin transcripts and peptides in various tissues reveals that penaeidins are constitutively synthesized and stored in the shrimp haemocytes. It was shown by immunocytochemistry, at both optical and ultrastructural levels, that the peptides are localized in granulocyte cytoplasmic granules, The expression and localization of penaeidins were further analysed in shrimp subjected to microbial challenge. Several outbreaks of gastroenteritis related to the consumption of shellfish (frequently eaten raw) have been reported in different parts of the world. In Europe, hum an calicivirus infections may have been involved in winter outbreaks in recent years, although there is little evidence confirming such viral contamination in shellfish. This study presents the first results of a field survey on viral contamination in two shelllish harvesting areas along the French Mediterranean coast. Progress in pathological characterization of Syndrome 85 in the black-lip pearl oyster Pinctada margaritifera ArchiMer

A programme of zoosanitary survey of the populations of black-lip pearl oyster (P. margaritifera) has been developed in French Polynesia during 1996 and 1997. Based on histological and ultrastructural examinations, the study of pearl oysters sampled in several pearl farms from the Society Islands, Tuamotu Atolls and Gambler Archipelago, has been particularly turned to enzootic pathological problems. Particular attention has been given to the shell disease and to the necrosis of the adductor muscle, symptoms that had accompanied the mass mortality in 1985.Virus-like particles, about 40 nm in diameter, were detected in membrane bound vesicles from the necrosized muscle tissue.
